From yesterday's Nintendo Direct live stream, Level 5 reveals the Professor Layton series will end with "Professor Layton Choubunmei to A no Isan."
While it wasn't revealed if the game will have a release outside of Japan in the future, the 6th and final game for the series will be for the 3DS and will end the Layton storyline.
Level 5 did tease the series itself might still be made in the future either as spin-offs or under a new playable character but the main series will end once the final game is released.
Level 5 also won't begin focusing on the final game until after "Professor Layton and the Miracle Mask" and "Professor Layton vs. Ace Attorney" are both released for the 3DS with the later set for a late 2012 release date.

The multiplayer was done by a separate studio.
